The image formats supported by AIJ are the same as the ones described here for base ImageJ:
http://rsbweb.nih.gov/ij/docs/guide/146-7.html

I am still not sure of the data format of your 32-bit tif image (RGB, 32-bit floating point, 32-bit integer). The first two are directly supported by ImageJ/AstroImageJ.

In any supported data format, the intensity measurement should be valid. RGB "sub-pixels" are combined into a single value for intensity calculations.

If you would like for me to examine your image to determine compatibility, please forward it directly to karenacollins ~~~at~~~ outlook ~~~dot~~~ com. In that case it will not show up here in the public forum.
